Membrane proteins need to be extracted from the lipid bilayer where they are embeded so that they are separated physically from each other for later purification. We usually use detergents that have amphipathic properties to extract membrane proteins from the cell membrane. The extraction of a membrane protein by a detergent consists of several stages. The detergent molecules first insert into the membrane bilayer, and then disrupt the membrane bilayer into pieces upon its increased concentration. Further increment of detergent concentration leads to the formation of the lipid/protein/detergent complex. For a specific membrane protein, the type of suitable detergent and the optimal molecular ratio of detergent: lipid: protein need to be found so that a membrane protein can be extracted in a natively folded state. Once the formulas for detergent, lipid, protein are determined, other factors, such as compatibility with protein detection and identification, and chromatography methods need to be considered.
