The transmembrane regions of multi-pass transmembrane proteins are highly hydrophobic, making it difficult to maintain the correct conformation in buffered aqueous solutions. Therefore, appropriate detergents and lipid molecules are needed to stabilize membrane proteins in their native conformations in the aqueous solutions. In addition, detergents are also crucial for extracting membrane proteins from the lipid bilayer of cell membranes.
