In order to extract membrane proteins, cells need to be disrupted by either ultrasound or high-pressure methods. The most common approach is to homogenize tissues or cells into an emulsified mixture in a buffered aqueous solution. A major concern during this process is that the target membrane proteins will be exposed to the proteases released from subcellular compartments and undergo proteolysis-caused degradation The proteolysis of the target proteins can be prevented or reduced by the addition of protease inhibitors.
